摘要
目的 研究凋亡相关基因bcl 2和抑癌基因 p16在食管鳞癌及癌旁组织中的表达及意义。 方法应用SP法 ,检测 4 0例食管鳞癌及癌旁组织中bcl 2和p16蛋白的表达。结果 bcl 2在正常食管粘膜、单纯增生、不典型增生及鳞癌组织中表达率分别为 6 %、6 3.3%、4 6 .5 %和 4 2 .5 % ,正常粘膜组与其余各组之间有显著性差异 (P <0 .0 1) ;bcl 2表达与病理分级、浸润深度及淋巴结转移无相关性。p16蛋白在正常食管粘膜上皮、单纯增生、不典型增生、鳞癌组织中的表达呈递减的趋势 (P <0 .0 1)。p16表达与病理分级、浸润深度及淋巴结转移相关。结论 bcl 2与 p16在食管鳞癌发生的不同时期起作用 ,bcl 2可作为食管癌发生的早期判断指标。p16蛋白的缺失与食管鳞癌的发生及恶性进展有关 ,检测 p16蛋白可能成为判断食管鳞癌预后的一个指标。
Objective To study the expression and significance of bcl-2 and p16 in esophageal squamous cell cancer and the surrounding tissues. Methods S-P method was used to examine the expression of bcl-2 and p16 in 40 esophageal squamous cell cancer and the surrounding tissues. Results Expression of bcl-2 was 6%, 63.3%, 46.5% and 42.5% in normal esophageal mucosa, atypical hyperplasia, and squamous carcinoma tissues, respedively. There were significant differences between normal esophageal mucosa and other groups (P< 0.01); The expression of bcl-2 had no relationship to the differentiation grade of carcinoma、invasion and lymph node metastasis (P> 0.05). p16 immunoreactivity decreased progressively from normal esophageal mucosa, simple hyperplasia, and atypical hyperplasia to squamous carcinoma tissues (P< 0.01). There were relationships in the expression of p16 and the differentiation grade of carcinoma、invasion and lymph node metastasis. Conclusion bcl-2 and p16 play roles in the different stage of occurrence of esophageal squamous cell cancer, bcl-2 may predicting the occurrence of esophageal squamous cell cancer in early stage. The loss of p16 protein expression may relate with the occurrence and progress esophageal squamous cell cancer and may be regarded as an useful index for prognosis.
