摘要
目的 制备抗重组人血管生长素 (recombinanthumanangiogenin ,rhANG)多克隆抗体 ,并初步应用于消化道肿瘤实验研究。方法 用重组基因工程菌表达 ,SDS法纯化的rhANG蛋白免疫新西兰兔 ,制备并纯化多克隆抗体 ,并用免疫组织化学和ELISA等方法对多克隆抗体进行测定。结果 此次获得的抗血清ELISA方法效价达 1∶12 80 0 ,检测ANG灵敏度最低为 8～ 16μg/L ,不与其他细胞因子和血清反应。该抗体与 2 0 %～ 3 0 %上消化道恶性肿瘤呈阳性反应。结论 ANG多克隆抗体对表达ANG的肿瘤组织反应特异性强 ,ELISA方法检测灵敏度高 。
Objective To prepare polyclonal antibody against recombinant human angiogenin (rhANG) and to study its application in the study of gastro intestine tumor. Methods\ New Zealand rabbit was with ANG protein expressed in E.coil with recombinant plasmid and purified by SDS. Polyclonal antiserum was purified and analyzed by ELISA (Enzyme linked immunsosrbent assay) and immunohisto chemical staining. Results\ The titer of the antiserum obtained in this experiment by ELISA was up to 1∶ 12 800 and the sensitivity was at least 8～16 μg/L. In addition, the antibody did not crossreact with other cytokines or serum. ANG immunostaining was positive in 20%～30% of upper digestive tract malignant tumors. Conclusion\ The specificity of polyclonal antibody against rhANG is high to tumor tissues that express ANG, and the method of ELISA is sensitive. This antibody can be used for detection of ANG.
