摘要
目的 :探讨 6 0 m(70 0 k Pa)模拟空气反复潜水对大鼠免疫功能的影响。 方法 :将大鼠置动物加压舱内 ,暴露于 70 0k Pa空气环境下 6 0 min,每天 2次 (8:0 0~ 9:0 0 ,14 :0 0~ 15 :0 0 ) ,连续 3d。于暴露后第 1、3、5天晨断头取血及脾脏。采用流式细胞术测定外周血及脾淋巴细胞亚型 ,用 3H- Td R掺入法测定脾淋巴细胞增殖反应 ,以 EL ISA法测定循环血浆及脾淋巴细胞经刀豆球蛋白 (Con A)刺激后分泌的 IL- 2水平以及比色法测定血浆中超氧化物歧化酶 (SOD)、过氧化氢酶 (CAT)、谷胱甘肽过氧化物酶 (GPx)、谷胱甘肽 S转移酶 (GST)的活力和还原型谷胱甘肽 (GSH)、丙二醛 (MDA)的含量。 结果 :经高压空气反复暴露后 ,外周血及脾 CD3+、CD4 + CD3+细胞含量及 CD4 + CD3+ / CD8+ CD3+比值均显著下降 (P<0 .0 5 ,P<0 .0 1) ,CD8+ CD3+ 细胞含量无明显变化 ;脾淋巴细胞增殖率显著下降 (P<0 .0 1) ;血浆及脾淋巴细胞经 Con A刺激后分泌的 IL - 2均显著下降 (P<0 .0 5 )。淋巴细胞亚型变化在反复暴露后第 3天即完全恢复正常 (P<0 .0 5 ,P<0 .0 1) ,暴露后第 5天仍维持于正常对照水平 ;脾淋巴细胞增殖率、血浆及脾淋巴细胞经 Con A刺激后分泌的 IL - 2水平在暴露后第 3天均上升 (P<0 .0 5 ) ,至第5天恢复至对照水?
Objective: To explore the immune function changes after 60 m repetitive air diving in rats. Methods: Adult rats were placed in an animal chamber(700 kPa air,for 60 min) twice a day(8:00 9:00, 14:00 15:00) for 3 d. Peripheral blood and spleen were sampled on the 1st, 3rd and 5th day following exposure. The subpopulations of lymphocyte in blood and spleen were identified using 3 coloured flow cytometry. 3H TdR incorporation were used to assess the proliferation rate of splenic lymphocyte. IL 2 in plasma or excreted by splenic lymphocytes after ConA stimulation were determined with ELISA. Activities of superoxide dismutase (SOD),catalase (CAT),glutathione peroxidase (GPx),glutathione S transferase (GST) and levels of glutathione (GSH),methylenedioxyamphetamine (MDA) in plasma were determined by colorimetry.Results: After hyperbaria exposure CD3 +, CD4 +CD3 + cells and the CD4 +CD3 +/CD8 +CD3 + ratio all decreased in both peripheral blood and spleen( P <0.05 and P <0.01). The percentage of CD8 +CD3 + cell didnot change. The proliferation rate of splenic lymphocyte and the IL 2 in plasma or excreted by splenic lymphocyte after ConA stimulation were all reduced ( P < 0.05 and P < 0.01). The changes in lymphocyte subpopulations all disappeared on the 3rd day after exposure ( P < 0.05) and remained normal on the 5th day. The proliferation rate of splenic lymphocyte and the IL 2 in plasma or excreted by splenic lymphocyte after ConA stimulation increased gradually on the 3rd day( P <0.05) and recovered to the control levels on the 5th day ( P < 0.01 and P <0.05). Conclusion:There exist inhibitory effects of 60 m simulated air repetitive diving on rat immune functions which can recover in 3 5 d.
