摘要
目的 探讨人精子膜上的整合素 VL A- 5是否参与了人精子的激活。方法 采用整合素 VL A - 5的特异性配体纤连蛋白 ( Fn)作用于生育组和不育组人精子 ,以三色法染色观察 Fn作用前后顶体反应 ( AR)率的变化 ;预先用 Fluo- 3 /AM负载人精子 ,用流式细胞仪检测 Fn作用前后精子 [Ca2 + ]i 荧光强度的变化。结果 生育组 10 0nmol/L的 Fn作用于人精子后 AR率由 8.64 %± 2 .5 6%增高到 16.0 9%± 3 .18% ( P<0 .0 1) ,钙离子的荧光强度值由静息状态的 5 .73± 2 .45升高到 6.61± 2 .40 ( P<0 .0 1)。不育组 Fn作用前后 AR率由 7.0 9%± 0 .94%增高到10 .44 %± 2 .3 0 % ( P<0 .0 5 ) ,钙离子的荧光强度值无显著改变。结论 人精子膜上的整合素 VL A -
Objective This study was aimed to disclose whether the VLA-5 integrin on human sperm membrane takes part in the activation of human spermatozoa. Methods Fluo-3/AM was coincubated with human spermatozoa. The change of intracellular calcium( i) was determined by flow cytometry when human sperms were exposed to Fibronectin(Fn) as the specific ligand of integrin VLA-5. Human sperm acrosome reaction(AR) rate was evaluated by triple-stain technique. Results Fn significantly raised the AR rate (P<0.05) and the level of i (P<0.01) in the spermatozoa of fertile men. But there was no significant rise in the level of i in the spermatozoa of infertile men. Conclusion The VLA-5 integrin on human sperm membrane plays a role in the activation of sperm.
