摘要
目的 比较海藻糖和蔗糖在人红细胞冰冻干燥保存过程中对细胞的保护效果。方法 将浓缩红细胞、含有不同浓度海藻糖或蔗糖的30%聚乙烯吡咯烷酮(PVP)按照1:3(V/V)的比例混合,-80℃冰箱中预冻1h,入冻干机内冻千处理后,用37℃等渗缓冲液快速水化洗涤样品,然后将其分为3个处理组:组1为30%PVP组(简称PVP组),组2、3为含有5%、10%和15%海藻糖或蔗糖的30%PVP组(简称海藻糖组或蔗糖组),测定各项指标。结果 冻干红细胞再水化后,海藻糖组的细胞回收率[(18.86±4.63)%、(21.73±7.32)%和(18.01±4.53)%]显著低于PVP组[(45.97±11.77)%](P<0.01);蔗糖组,蔗糖浓度为5%、10%时,细胞回收率分别为(37.40±2.90)%和(37.77±4.78)%,二者显著低于PVP组(P<0.05),但当蔗糖浓度升至15%时,细胞回收率[(42.54±10.25)%]和PVP组无显著差异;血红蛋白回收率,海藻糖组[(50.00±3.47)%、(40.91±9.09)%和(52.09±7.01)%]显著低于PVP组[(77.27±3.63)%](P<0.05),而蔗糖组和PVP组无显著差异,且显著高于海藻糖组(P<0.05)。洗涤后3组的游离血红蛋白浓度均<1g/L,其余各项指标的对比关系类似于水化后指标。结论 在冻干保护液中添加不同浓度的海藻糖或蔗糖对于提高冰冻干燥保存后红细胞的回收率和血红蛋白浓度作用不明显;
Objective To compare the protective effect of trehalose with that of sucrose in lyophilization of red blood cells. Methods Concentrated red blood cells and 30%PVP containing different concentrations (5% ,10% and 15%) of trehalose (or sucrose) were mixed at a ratio of 1 : 3, pre-frozen at - 80℃ for 1h.then lyophilized in freeze-dryer. After lyophilization, the samples were reconstituted and washed in 37℃ isotonic buffer. The experiment was divided into 3 groups: 30%PVP group,trehalose group (containing 5%, 10% and 15% trehalose) and sucrose group (containing 5% , 10% or 15% sucrose). Results After lyophilization and rehydration, the recovery of red blood cells in trehalose groups were (18.86 ±4.63) %, (21.73 ± 7.32) % and (18.01±4.53) % respectively and significantly lower than that of PVP group[(45.97+11.77)%](P<0.01).The recovery of red blood cells were (37.40 ± 2.90)% and(37. 77 ± 4. 78) % and were lower than the PVP group (P < 0.05) at sucrose concentrations of 5 % and 10 % . The recovery of red blood cells [(42.54±10.25)%]was not significantly different from the PVP group at 15% sucrose concentration. The recovery of hemoglobin in trehalose groups were (50.00 ± 3. 47) % , (40. 91 ± 9. 09) % and (52. 09 ± 7. 01) % respectively, which were significantly lower than the PVP groupt (77.27 ± 3.63) % ] (P < 0.05). The recovery of hemoglobin of the sucrose groups were(72.73 ± 8.31)%,(70.31 ± 11. 27)%,and(72. 73±4.97)% respectively, which were similar to the PVP group,but significantly higher than the trehalose groups(P<0. 05) .After washing,the concentration of free hemoglobin in all groups were lower than 1g/L. Conclusion Addition of trehalose or sucrose in protective solutions can not significantly increase the recovery of human red blood cells and hemoglobin after lyophilization; sucrose can increase DImax of red blood cells after lyophilization,so the effect of sucrose in lyophilization is better than that of trehalose.
