摘要
目的 :探讨二甲基亚砜 (DMSO)对颅脑损伤后细胞凋亡调控基因Bcl- 2变化的影响。方法 :采用自由落体撞击脑损伤模型造成中度颅脑损伤 ,治疗组伤后经尾静脉给予DMSO 5 0mg/kg。损伤对照组、治疗组分别于损伤后 6h、1 2h、2 4h、48h、72h和 1 68h随机点杀动物 6只 ,空白对照组于 6小时一次处死。采用免疫组织化学方法检测凋亡基因Bcl- 2表达情况。结果 :治疗组Bcl - 2蛋白表达平均阳性细胞数明显增加 ,与损伤对照组对应时相比较P <0 .0 1。结论 :DMSO的应用可以阻止抗凋亡基因Bcl- 2的下调 ,从而抑制脑损伤后所致的细胞凋亡。
Objectives: To investigate the effects of the dimethyl sulfoxide on the varieties of the gene of Bcl-2 after traumatic brain injury. Methods: Both injuries and treatment with dimethyl sulfoxide were given to the trial group. The control group only received injuries and neither was given to the normal group. The free-falling-body impact device was used to cause local moderate contusion and laceration in the rats. Every rat of the trial group was given dimethyl sulfoxide (50mg/kg) by venipuncture after traumatic brain injury. Rats of the control group and the trial group were killed at 6h 12h 24h 48h 72h 168h after injury respectively. All of the rats from the normal group were killed at 6h after injury. The expression of Bcl-2 was evaluated by immunohistochemical detection. Results: The average positive cells expressing Bcl-2 protein in the trial group are obviously higher than that of the control group (p<0.01). Conclusion: Dimethyl sulfoxide can prevent Bcl-2 from downregulation and apoptosis after traumatic brain injury.
