猪瘟病毒的免疫酶组织化学定位

A COMPARATIVE STUDY OF IMMUNOCHEMISTRY FOR DETECTION AND LOCALIZATION OF SWINE CHOLERA ANTIGEN IN TISSUE

摘要 本实验结果证实PAP法在检测、定位猪瘟病猪组织中的病毒抗原是一种特异性强、敏感性高的一种新技术。主要优点是:①背景浅,阳性显色好,色深清楚;②适于检测、定位微量抗原。为病理诊断和鉴别诊断提供了可靠的形态学和免疫学依据。 The unlabeled antibody enzyme method of immunohistochemistry (PAP method) which has developed in the recent years is more sensitive than enzyme labeled antibody technique. The methods for preparation of rabbit antihorseadish peroxidase (anti-HRP) sera and soluble complex (PAP) were recommended. The formula for calculating the ratio of HRP to anti-HRP in PAP was given.12 cases of swine cholera pigs, 2 cases of lappinized weak virus vaccinated pigs and 2 cases of non-vaccinated strong pigs with 54 formalin fixed paraffin cmbeded blocks in all were examined for detection and localization of swine cholera antigen by direct, indirect enzyme labeled method and PAP method. Comparing the results of these methods used, the PAP method was proved more sensitive and therefore may be used in the localization of virus antigen and pathologic diagnosis of swine cholera.
