盐胁迫对胡杨液泡膜H^+ATPase水解活性的影响 被引量:10

摘要 胡杨细胞经 50mmol·L- 1 NaCl处理 1 0d后 ,泡膜液H+ ATPase,PPiase水解活性都增加了 ,说明胡杨具有盐生植物的特性。盐胁迫对专一性抑制剂bafilomycinA1 ,DCCD更敏感 ,说明盐处理下胡杨液泡膜H+ ATPase与bafilomycinA1 ,DCCD结合位点上的构象可能发生了一定的变化。从Mg2 + ,ATP依赖性 ,NO3- 和bafilomycinA1 抑制特性、Cl- 激活特性证明胡杨液泡膜H+ ATPase是典型的V H+ ATPase。盐胁迫后 ,对底物ATP的亲和力增强。 Populus euphratica suspension-cultured cells were t reated with 50 mmol·L -1 NaCl for 10 days, the H +-ATPase and H +-PPia se hydrolytic activity were both increased.The H +-ATPase was more sensitive to specific inhibitor Bafilomycin A 1 and DCCD, it might change its conformatio n under salt stress. Mg 2+ , ATP-dependent, Cl - stimulated, specific inhi bitor NO - 3 and bafilomycin A 1 indicated the H +ATPase in the tonoplast of P.euphratica belonged to vacuolar type ATPase. It had greater affinity for ATP than the control.
