摘要
采用改良CTAB法、改良SDS法、快速提取法分别以玉米种子、黄化苗叶片、正常生长期绿色叶片为材料提取细胞总DNA。结果表明用 3种不同方法提取的DNA纯度符合分子生物学实验要求。不同部位的材料、不同的提取方法 ,DNA得率不同。玉米干种子优适用于在快速提取法中提取DNA ;改良CTAB法、SDS法宜从玉米黄化苗叶片和正常绿色叶片中提取DNA。
Total maize DNA was respectively extracted from seeds,yellow seedling,green leaf with 3 kinds of extraction DNA methods. The result showed the DNA extracted using 3 kinds of methods accorded with demand of the quality standard of DNA in molecular experiment.The extraction rates of DNA were variable with different material and extraction methods. Seeds are the best material for rapid extraction method.Gaining plenty of DNA from the yellow seedling and green leaf was best used for Modified CTAB and SDS.
