摘要
采用一种改进的简单制样方法 ,垂直板电泳 ,对采自我国不同地区的 31个根结线虫群体的几种同工酶进行了分析。通过分析 ,发现 5个群体是混合群体 ;在含有 8个种的 31个群体中获得 8种酯酶表型 ,包括一种未报道类型 ,即Rf值为 0 .440和 0 .480的S1 -M 1a型 ;在 7种 2 2个群体中测出 5种苹果酸脱氢酶表型 ,包括两种未报道类型 ,即Rf值为 0 .2 90的N1b型和为 0 .2 2 8和 0 .2 90的N2a型 ;在 6种 2 0个群体中测出 6种超氧化物歧化酶表型 ,包括两种未报道类型 ,即Rf为 0 .60 8和 0 .643的I2a型和为 0 .570的E1型 ;在 6种 1 9个群体中仅测出 2种过氧化氢酶表型 ,可能受寄主植物的影响 ;在 5种 1 8个群体中测出与寄主植物种类密切相关的 2种过氧化物酶表型。研究结果表明 ,根结线虫种类鉴定中 ,酯酶最有价值 ,苹果酸脱氢酶和超氧化物歧化酶次之 ,过氧化氢酶可能没有价值 。
Five isozymes from 31 Meloidogyne populations from China were ana lyzed by electr ophoresis in 1.5-mm-thick polyacrylamide gels. Among 31 populations of 8 species ,12 major bands of esterase(EST) activity were detected, corresponding to 8 distinct phenotypes, including an uncovered o ne, i.e. S1-M1a with Rf= 0.440 and 0.48 0,which belonged to an unidentified population from Qingdao. Among 22 population s of 7 species, 4 bands of malate dehydrogenase(MDH) activity were detected, cor responding to 5 different phenotypes, including two uncovered ones,i.e.N1b with Rf=0.290 and N2a with Rf=0.228 and 0.290, which belonged to the unidentified p opulation from Qingdao and M.enterolobii,respectively. Among 20 po pulations of 6 species, 8 major bands of superoxide dismutase(SOD) activity were detected, cor responding 6 distinct phenotypes, including two uncovered ones, i.e. E1 with Rf= 0.570 and I2a with Rf=0.608 and 0.643, which belonged to M. enterolobii and the unidentified population from Qingdao, respectively. Among 19 populations of 6 sp ecies, only 2 distinct catalase(CAT) phenotypes were detected and they seemed to be affected by the host plants. Among 18 populations of 5 species, the detected PO phenotypes were obviously related to the host plants. According to the resul ts, in the identification of Meloidogyne species, EST was of the m ost value, MDH and SOD were less useful but very helpful in combination with the use of EST, C AT was probably useless and PO had no value.
