摘要
目的探讨吸入外源性一氧化氮 (nitricoxide,NO)对正常大鼠肾脏的毒副作用。方法 72只大鼠随机分为三组 ,均行气管切开 ,插气管导管。吸氧组 (G1 )单纯吸氧 (FiO2 0 .4 0 ) ,低浓度NO吸入组 (G2 )和高浓度NO吸入组(G3 )除吸氧外 ,分别吸入 4 0ppm和 80 ppm的NO ,于 2、8、1 2、2 4h取标本检测 ;另取 6只大鼠建立正常值和组织学对照。数据行多个样本均数间方差分析。结果三组肾脏含水量无明显差异 (P >0 .0 5 ) ,三组呼出气NO2 浓度G3 组比G1 和G2 组显著增高 (P <0 .0 5、P <0 .0 1 ) ;三组Cr变化不显著 (P >0 .0 5 ) ,G3 组血尿素氮和高铁血红蛋白分别高于G1 和G2 组 (P >0 .0 5、P <0 .0 1 ) ,G1 和G3 组均出现肾脏组织形态和超微结构的病理性变化 ,但G2 组病理变化较轻。结论吸入低浓度NO对肾脏无明显毒副作用 ,吸入 80
ObjectiveTo evaluate the toxicity and side effects of exogenous nitric oxide (NO) on the kidney of rats.Methods72 rats were divided into three groups,Group1 (G 1) inhaled O 2 (FiO 2= 0.40 );Group2 (G 2) inhaled O 2 (FiO 2= 0.40 )+40ppm NO;Group3 (G 3,n=24) inhaled O 2 (FiO 2= 0.40 )+80ppm NO; After 2,8,12 and 24 hours of the experiment,the specimen was examined.In addition,another 6 rats were only for histological comparison and to set up the normal value (n=6).The data was analyzed using ANOVA for multiple mean comparison.ResultsThere was not significantly different in the water content of the renal tissue among the three groups (P> 0.05 ).The concentration of NO 2 of G 3 was significantly higher than that of G 1 and G 2 (P< 0.05 and P< 0.01 ).The difference of creatinine(Cr) among the three groups were not significant(P> 0.05 ).Both the blood uria nitrogen(BUN) and methemoglobin(MHB) of G 3 were higher than that of G 1 and G 2(P< 0.05 ,P< 0.01 ).Pathological changes of kidney in G 2 was lighter than those in G 1 and G 3.ConclusionInhaling lower concentration NO had no significant toxicity or side effects to the rat's renal tissue,while inhaling 80ppm NO could impair rat's kidney in various degrees.
