摘要
目的 :通过对犬下颌骨牵张成骨过程中咀嚼肌肌球蛋白重链亚型mRAN含量变化的测定 ,研究牵张成骨过程中肌肉改建的过程及能力。方法 :12只西安本地杂种犬分为 6组 :对照组、牵张 5d组、牵张 10d组、固定 2周组、固定 4周组和固定 8周组。麻醉后分别取嚼肌与颏舌骨肌的中央部分对MHCⅠ、MHCⅡ及GAPDH(内参照 )的mRNA进行RT -PCR。结果 :试验各组嚼肌中各项检测指标与对照组相比没有显著性差异。牵张 5d后颏舌骨肌中MHCⅠ的mRNA相对含量开始上升 ,MHCⅡ的mRNA相对含量开始下降 ,MHCⅠ /MHCⅡ之比增加 ,刚刚牵张完成时变化最为明显 ,牵张完成固定 4~ 8周时基本恢复。结论 :在犬下颌骨牵张成骨过程中相关的咀嚼机会发生组份的改变 。
Objective:To study the ability and pattern of muscle remodeling about the relative contents of myosin heavy-chain isoforms mRNA in muscles of mastication during distraction osteogenesis of mandible in canine.Methods:Bilateral distraction osteogenesis was performed on 12 mongrel dogs,5 to 6 months of age.Animals were divided into 6 groups:control group,5th,10th day in distraction and 2nd,4th,8th week after distraction.After anaesthesia the central of masseters and geniohyoid were immediately dissected out and stored at -75℃,subsequently the mRNAs of MHCⅠ,MHCⅡ and GAPDH(home-keeping gene) were measured by RT-PCR.Results:In geniohyoid muscle,the amount of MHCⅠmRNA was significantly increased at 5th,10th day in distraction and 2nd week after distraction,the amount of MHCⅡmRNA was significantly decreased at 10th day in distraction,but the amount of MHCⅠ/MHCⅡwas significantly increased at 5th,10th day in distraction and 2nd week after distraction.In masseter there was no significant difference in all of observing result between distracted group and control group.Conclusions:Mandibular distraction osteogenesis of the canine induced the remodeling of associated masticatory muscles which improved long-term stability of mandibular expansion by distraction osteogenesis.
