摘要
目的 探讨Ⅰ型胶原对单核细胞摄取ox-LDL能力的影响。方法 采用密度梯度离心、贴壁粘附法分离冠心病和健康人外周血单核细胞,比较不同浓度Ⅰ型胶原及其降解产物对原代培养单核细胞摄取FITC标记ox-LDL能力的影响。结果 ①Ⅰ型胶原0、0.01、0.1、0.5g·L^(-1)作用下,ox-LDL(80g·L^(-1)同单核细胞孵育24h后ox-LDL结合摄取值分别为(2438.48±447.20)mg·g^(-1)、(2568.69±543.74)g·L^(-1)、(4972.40±814.30)mg·g^(-1)和(5747.90±416.14)mg·g^(-1)。其中,0.5、0.1g·L^(-1)Ⅰ型胶原同低浓度组相比具有显著性差异(P<0.001和P<0.05)。②ox-LDL(80g·L^(-1))孵育24h后,对照组、Ⅰ型胶原组及其降解产物组单核细胞ox-LDL的结合摄取值分别为(2554.07±550.71)mg·g^(-1)、(3403.27±442.08)mg·g^(-1)和(5304.73±577.87)mg·g^(-1),具有显著性差异(P<0.001)。结论 Ⅰ型胶原能以剂量依赖性方式显著提高单核细胞摄取ox-LDL,其降解产物的作用更为强大。
Objective To investigate the effect of type I collagen on ox-LDLs uptaken by mono-cytes.Methods By exsanguinating the venous blood from patients with CHO and normal people on an empty stomach and isolating monocyte from the blood by density gradient centrifugation using lymphocyte separation solution,we study the effect of type I collagen as well as its degradation products on uptaking ox-LDLs labeled with fluorescein isothiacyanate(FITC)by monocytes. Results (1) Monocyte was significantly increased uptake of ox-LDL(80 ug/ml) in a dose-dependent manner after incubating with type I collagen for 24 hours(P < 0.05) . (2) Monocytes in EDTA + type I collagen group and type I collagen group uptake more ox-LDLs than monocytes in plastic group( P < 0.05 and P < 0.001). Monocytes in type I collagen group uptake more ox-LDLs than monocytes in EDTA + type I collagen group(P < 0.001) .Conclusions Type I collagens increase ox-LDL uptaken by monocytes in a dose-dependent manner and its degradation products play more important role.
