摘要
对重组基因工程酵母 (PP NP 1)进行诱导培养 ,然后对诱导表达物进行饱和硫酸铵分级沉淀 ,再对沉淀进行Western印迹 ,证实其为特异的植酸酶 ;再对基因工程菌所产植酸酶进行纯化 ,并对其酶学性质进行了研究。研究结果 :测得其分子量为 70 2kD ;酶作用最适温度为 5 5℃ ;在 pH 2 5~ 5 6范围内植酸酶均具较高的酶活性 ,最适pH为 4 6 ;另外Ca2 + 、牛血清白蛋白、Mn2 + 、Mg2 + 对酶有激活作用 ,而HPO42 、SDS、Cu2 + 、Fe2 + 对酶有抑制作用。
