摘要
目的:用构建的细粒棘球蚴 95 (Eg95 )抗原基因真核表达重组质粒 pc DNA3- Eg95 ,直接免疫小鼠 ,观察其所诱导的体液免疫反应。方法:碱裂解法大量提取重组质粒 ,采用肌肉注射、静脉注射、皮下注射 3种免疫途径和不同剂量的重组质粒免疫小鼠 ,用 EL ISA法测定小鼠血清抗体滴度。 结果:在相同剂量下 ,重组质粒免疫小鼠刺激机体产生抗体反应强度的免疫途径依次为肌肉、皮下和静脉注射 ,阳性反应率由高到低依次为皮下、肌肉和静脉注射。以肌肉注射方式免疫小鼠的抗体反应维持时间较长 ,主要产生 Ig G2 a为主的抗体。 结论 :用 pc DNA3-Eg95重组质粒
Objective: To study the humoral immune responses in BALB/c mice vaccinated with the constructed Eukaryotic expression plasmid, pcDNA3, carrying Eg95 antigen gene from the Echinococcus granulosus. Methods: The constructed recombinant plasmid, pcDNA3 Eg95, were obtained by alkaline lysis and were injected into mice by the various doses and routes. The changes of anti Eg95 antibodies in the sera of mice were tested by ELISA. Results: It was found that the routes of plasmid pcDNA3 Eg95 inducing highly level of antibody response were intramuscular, intravenous and subcutaneous injections in turn. There was dependence to vaccine doses in mice. Conclusion: Vaccination of mice with the recombination plasmid, pcDNA3 Eg95,could elicit humoral immune response in mice.
