摘要 目的 建立反相高效液相色谱法测定人血清中茶碱浓度。 方法  采用高效液相色谱法 ,苯巴比妥为内标 ,经 YWG-C1 8色谱柱 ( 2 0 0 nm×4.6mm,10 μm)分离 ,流动相为甲醇∶水 ( 3 0∶ 70 ,V/V) ,流速为 1.0 ml· min- 1 ,柱温 3 5℃ ,2 10 nm波长处二极管阵列检测器检测。结果  茶碱血清浓度在 1.0 0~ 3 0 .0 0μg· ml- 1 浓度范围线性关系良好 ,r=0 .9994,血清最低检测限为 2 0 0 ng· ml- 1 ;平均回收率为 98.3 6% ;日内和日间 RSD分别为 2 .5 2 %和 3 .41%。 结论  本法灵敏简便、准确可靠 ,可用于茶碱血药浓度测定及其药代动力学研究。 OBJECTIVE To estalish a method of revered phase high performance liquid chromatography(RP HPLC)for the determination of theophylline concentration in human plasma.METHODS RP HPLC was carried out on YWG C 18 (200nm×4 6mm,10μm)..Phenobarbital was used as the internal standard.The mobile phase was composed of methanol water(30∶70,V/V),the flow rate was 1 0ml·min -1 ,the temperation of column was at 35℃,the detecting wavelength was 210nm.RESULTS The calibration curve was linear within the rangs from 1 00 to 30 00μg·ml -1 for theophylline,r=0 9994,and the lowest plasma limitated concentration was 200ng·ml -1 .The average recovery was 98 36%(n=9).The RSD within a day and between days were 2. 52% and 3 41%.CONCLUSION The method is sensitive,simple,rapid and accurate.It may be used for the determination of theophylline concentration in human plasma and its pharmacokinetic study.
