摘要
目的 了解高效液相色谱 (highperformanceliquidchromatography,HPLC)技术在兔直流电盐酸川芎嗪注射液离子导入后在测定玻璃体盐酸川芎嗪含量方面的应用。方法 色谱条件是色谱柱为ODSC1 8不锈钢柱 (1 50mm× 4 6mm ,5μm) ;流动相为甲醇∶水 =40∶60 ,1 0 0 0ml中含十二烷基磺酸钠 (SDS) 1g ,磷酸二氢钾 0 6g,冰醋酸 1ml;流速为 1 0ml·min- 1 ;检测波长为2 80nm ;室温操作。结果 在 0 54~ 1 7 2 8μg·ml- 1 范围内 ,峰面积与浓度呈良好线性关系 ,r =0 9994;最低检测浓度为 0 0 8μg·ml- 1 ;平均回收率为 92 542 % ;日内和日间精密度均小于 1 0 %。结论 建立专属、简单、快速和精确的高效液相色谱法可用于对盐酸川芎嗪在兔玻璃体内药代动力学的研究。
OBJECTIVE To assess application of application of high performance liquid chromatography(HPLC)for determination of hydrochloric ligustrazine in rabbit vitreous after transmitting the ion of injection of ligustrazine into vitreous. METHODS The analytical column was a ODS C 18 stainless steel column(150mm×4.6mm, 5μm). The mobile phase consists of 40 percent methanol and 60 percent H 2O. In 1000ml mixture of the mobile phase there are 1.0g SDS, 0.6g KH 2PO 4 and 1ml glacial acetic acid.The flow rate is 1.0ml·min -1 . The wavelength of the detector is 280nm. The operation is carried in room temperature.RESULTS The relationship between the peak area and concentration of the drug is linear within the range of 0.54~17.28μg·ml -1 with a correlation,r=0.9994. The minimal detectable limit was 0.08μg·ml -1 . The average recovery is 92.542 percent. RSD of intra-day and inter-day are all lower than 10%. CONCLUSION HPLC is a specific,simple,rapid,accurate method, and the suitable method established could be applied for the pharmacokinetic research of hydrochloric ligustrazine in rabbit vitreous.
