热电厂粉尘对大鼠支气管肺泡灌洗液成分及肺组织细胞因子释放的研究 被引量:3

摘要 目的 探讨尘肺的生物学标志物及不同粉尘致病力不同的机制。方法 收集尘肺患病率明显不同的甲、乙两热电厂粉尘 ,以生理盐水作为阴性对照 ,石英尘作为阳性对照 ,分别对大鼠行一次性气管内注入染尘。染尘后 10d ,观察不同粉尘引起大鼠早期支气管肺泡灌洗液 (BALF)中乳酸脱氢酶 (LDH )、总蛋白、磷脂及肿瘤坏死因子 α(TNF α)含量的变化 ,免疫组织化学染色观察肺组织TNF α的表达。结果 石英尘、甲、乙两厂粉尘均引起大鼠BALF中LDH、总蛋白含量增加 ,与对照组比较差异具有显著性 ,按大小顺序排列为石英组 >甲厂组 >乙厂组 (P <0 0 5 )。石英尘和甲厂粉尘导致大鼠BALF中磷脂含量比对照组明显增加 ,石英组高于甲厂组 (P <0 0 5 ) ,乙厂粉尘BALF中磷脂含量与对照组比差异无显著性 (P >0 0 5 )。三种粉尘均引起BALF中TNF α免疫活性增强 ,按大小顺序排列为石英组 >甲厂组 >乙厂组 (P<0 0 5 )。从乙厂粉尘到甲厂粉尘到石英尘 ,引起大鼠肺组织TNF α阳性表达逐渐增强 (P <0 0 5 )。结论 染尘大鼠早期BALF中增高的LDH活性、总蛋白和磷脂的含量及肺组织中TNF α的表达量与粉尘的致病潜力一致。 Objective To elucidate the biological marker of pneumoconiosis and the pathogenetic mechanism induced by different dusts. Methods The rats were injected intratracheally with quartz and the dusts from A and B two power stations respectively. The incidence of pneumoconiosis in power station A was significantly higher than that in B. Controls were similarly injected with physiological saline. All the animals were killed 10 days after dust injection. The lungs were immediately lavaged. The cell-free supernatant was separated for examining the contents of lactate dehydrogenase (LDH), the total proteins, the total phospholipid and tumor necrosis factor-α(TNF-α). The lung sections were placed on slides for immunohistochemistry of TNF-α. Results In comparison to those of controls, the activity of LDH and the total proteins were significantly elevated by all three kinds of dusts. The degree of increase was of the order: quartz>dust from A>dust from B( P <0.05). The total phospholipid contents in the quartz group and the dust from A group was significantly higher than that in the controls, and the former was higher than that of the latter( P <0.05). However, the dust from group B didn't show the change( P <0.05). A significant increase of the TNF-α release in brochoalveolar lavage fluid (BALF) and the lung tissue was observed in the following order: quartz>dust from A>dust from B.Conclusion The results demonstrated that elevated levels of LDH, the total protein, the total phospholipid and the TNF-α expression appeared to be paralleled with the degree of the known fibrogenic potential of dusts.
