大鼠短暂性局灶性脑缺血后侧脑室室下带神经细胞再生的观察 被引量:3

The Observation of Neurogenesis in the Subventricular Zone in Rats after Transient Fo- cal Cerebral Ischemia

摘要 目的:观察大鼠短暂性局灶性脑缺血后侧脑室嘴侧的室下带(SVZ)神经细胞再生的时间规律。方法:行线栓法制作局灶性脑缺血模型,用BrdU标记再生的神经细胞,再用免疫组织化学SP法检测含BrdU的细胞。观察短暂性局灶性脑缺血后1、4、7、10、14天SVZ神经细胞再生的数量。结果:缺血后4天BrdU阳性细胞明显增加,7天时达到峰值,随后开始下降,在14天时明显下降,但仍高于正常组。结论:短暂性局灶性脑缺血可促进室下带的神经细胞再生,提示脑有自我修复的潜在能力。 Objective: To observe the regularity of transient focal cerebral ischemia and neurogenesis of subventricular zone. Methods: Use middle cerebral artery occlusion (MCAO)to make focal cerebral ischemia model. BrdU, the thymidine analog that can incorporated into the DNA during the S phase of the cell cycle. Investigate the numbers of neurogenesis in the SVZ at 1, 4, 7, 10, 14 days after transient focal cerebral ischemia. Results: There was a significant increase in the numbers of BrdU immunoreactive cells in sub-ventricular zone at 4 days and with a peak at 7 days, then the numbers started to decease, but were still more than normal. Conclusion: Transient focal cerebral ischemia may enhance the neurogenesis of the subventricular zone and suggest that the cerebral has self-renovated capacity.
