鲍曼不动杆菌SHV-12型超广谱β内酰胺酶基因研究 被引量:6

摘要 目的 分析自临床分离的产超广谱 β 内酰胺酶 (ESBLs)鲍曼不动杆菌SHV型 β 内酰胺酶耐药基因序列 ,并确定其所产ESBLs亚型。 方法 从住院病人的送检标本中分离的 6 0株鲍曼不动杆菌中 ,经药敏试验、ESBLs表型确证试验和SHV型 β内酰胺酶耐药基因的聚合酶链反应 (PCR)检测 ,筛选出 1株具有多重耐药特性、ESBLs表型和SHV基因型均阳性的分离株 (HZ0 2株 ) ,对其耐药基因进行序列分析。 结果 HZ0 2株基因片段长 82 5个核苷酸 ,与Nuesch -Inderbinen等从肠杆菌中发现的SHV 12型ESBLs编码基因序列 (GenBank注册号 :X9810 5 )完全相同。 结论 HZ0 2株鲍曼不动杆菌可产生SHV 12型ESBLs,从而证实了我国临床分离的鲍曼不动杆菌中存在产SHV 12亚型ESBLs菌株。其序列已在美国核酸数据库 (GenBank)登录 (注册号 :AY2 5 916 3)。 Objective To investigate SHV type β lactamases encoding genes sequence and identify its subtype of multiple drug resistant Acinetobacter baumannii producing extended spectrum β lactamases(ESBLs) isolated from Huzhou District, Zhejiang Province, China. Methods Sixty strains of clinically isolated Acinetobacter baumannii were collected from July 2000 to December 2002, antibiotic sensitive test and confirmatory test for ESBLs were performed by agar dilution method.SHV type β lactamases encoding genes were tested by polymerase chain reaction (PCR).SHV sequence of Acinetobacter baumannii HZ02 strain was detected by ABI automated sequencer and was compared with SHV genes that had been published in Gen Bank. Results Eight hundred and twenty five nucleotides were determined by detecting SHV sequence of Acinetobacter baumannii HZ02 strain. It had the same gene sequence as the gene encoding SHV 12 subtype of ESBLs discovering from Enterobacteriaceae in Swiss (Gen Bank Accession No.X98105). Conclusion Acientobacter baumannii HZ02 strain may produce SHV 12 subtype of ESBLs gene. SHV sequence of Acinetobacter baumannii HZ02 strain has been registered in GenBank (Accession No.AY259613).
