摘要
目的 :从细胞凋亡角度研究六神丸及蟾酥、雄黄治疗白血病的机制。方法 :MTT法观察六神丸对HL 6 0白血病细胞的生长抑制、用荧光、瑞士 吉姆萨染色法观察细胞凋亡的形态学变化 ,RT PCR方法检测相关基因Bcl 2、C myc、BaxmRNA表达。结果 :六神丸及蟾酥水溶液有明显促进白血病细胞HL 6 0凋亡的作用 ,而含同样剂量雄黄的水溶液未见明显促凋亡作用 ,珍珠、牛黄、麝香、冰片混合物的水溶液也无明显促凋亡作用。六神丸能下调凋亡相关基因Bcl 2、C myc、mRNA表达水平 ,上调BaxmRNA表达水平。结论 :六神丸可诱导白血病细胞凋亡 。
Objective :To investigate the mechanism of Liu Shen Wan,vallicepobufagin and realgar in treating leukemia from the aspect of apoptosis. Methods :MTT assay was used to observe the growth inhibition on HL 60 cells of Liu Shen Wan,vallicepobufagin and realgar,the fluorescence and Wright Giemsa dying assay were used to observe the morphologic changes of apoptosis.And the molecullar assay of RT PCR was used to examine the mRNA level of gene bcl 2,c myc and bax. Results :The water solution distilled from Liu Shen Wan and vallicepobufagin had obvious effects on inducing HL 60 apoptosis,while realgar had no such effect.The mixture of Peral,bezoar,musk and borneol had no effects of inducing apoptosis,either. Conclusion :Leukemia cell can be induced apoptosis by Liu Shen Wan and Liu Shen Wan is an effective prescription in treating leukemia.
