应用原位末端转移酶标记和DNA电泳检测白内障晶体上皮细胞凋亡

摘要 目的:探讨老年性白内障晶体上皮细胞凋亡与白内障发生的关系。方法:应用原位末端转移酶标记和DNA片段凝胶电泳法,对50例老年性白内障和20例正常透明晶体上皮细胞进行凋亡细胞检测。结果:用Tunel法检测老年性白内障晶体上皮细胞,凋亡细胞比例为8.4%~37.8%,而正常透明的晶体上皮细胞中凋亡细胞百分率≤0.01%,两组有统计学差异;琼脂糖凝胶电泳结果显示,在老年性白内障组中出现了典型的DNA梯状条带,而正常组晶体上皮细胞DNA无此现象。结论:晶体上皮细胞凋亡与老年性白内障的发生发展有关。 Objective:To study the relation between senile cataract and apoptotic lens epithelial cells.Methods:The lens epithelial cells from50cases of senile cataract and20cases of transparent lens were detected by the methods of Tunel(Terminal deoxynucleltidyl transferase mediated duTp nick and Label Ling assay)and DNA Agarose gel electrophoresis.Results:The apoptoic cell rate was8.4%~37.8%in senile cataract.DNA products extracted from the cells of senile cataractous lenses manifested a typical DNA ladder after agarose gel electrophoresis,while those from the normal lenses showed no DNA ladder.Conclusion:The formation of senile cataract should be related to apoptosis of lens epithelial cells.
