TRAIL基因在白血病原代细胞中的表达

摘要 目的:检测白血病原代细胞和正常人外周血淋巴细胞中TRAIL基因的表达情况,探讨TRAIL凋亡途径在肿瘤免疫中的作用。方法:通过RT-PCR检测TRAIL基因的表达。结果:83%(10/12)淋巴系白血病和80%(16/20)髓系白血病患者TRAIL基因表达阳性,正常人外周血淋巴细胞TRAIL基因表达阴性,IL-2能诱导PBL的TRAIL基因表达。结论:大多数白血病原代细胞TRAIL基因表达阳性,可能在肿瘤的自发坏死和凋亡以及免疫逃逸中发挥作用。另外,活化的T细胞表达TRAIL基因,TRAIL可能参与CD95非依赖性激活诱导的细胞凋亡。 Objective:To determine the expression of TRAIL gene in primary leukemic cells and PBLs from normal individuals and study its possible role in tumor immunology.Methods:RT-PCR was used to determine TRAIL gene expression.Results:TRAILmRNA transcripts were detected in10/12lymphocytic leukemia and16/20myelocytic leukemia samples by RT-PCR.There was no detectable TRAIL mRNA in PBLs from normal individuals,and IL-2could induce its expression.Conclusion:Most primary leukemic cells expressed TRAIL,suggesting TRAIL might play a role in tumor cell evasion of immunosurveillance and spontaneous tumor cell death and necrosis.In addition,expression of TRAIL was found in activated T cells,suggesting that TRAIL might participate in CD95-independent apoptosis of lymphoid cells.
