摘要 目的:探讨CD-TK融合自杀基因体系对前列腺癌PC-3m细胞的杀伤作用。方法:应用逆转录病毒载体将CD-TK融合基因转染PC-3m细胞,经RT-PCR鉴定后,用MTT法检测丙氧鸟苷(GCV)、5-胞嘧啶(5-FC)前体药物单一或联合应用对转染PC-3m细胞的杀伤作用,以未转染PC-3m细胞为对照。同时应用流式细胞仪、电镜等检测药物作用前后的变化。结果:GCV对转染PC-3m细胞有较强的直接细胞毒作用,而5-FC治疗有显著旁观者效应;联合用药具有协同作用。结论:CD-TK融合基因治疗对前列腺癌细胞具有显著杀伤作用,明显优于单一自杀基因治疗,深入研究意义较大。 Objective:To investigate the effect of CD-TK fusion gene therapy on human prostate cancer.Methods:MTT assay,flow cytometry(FCM),and electrical microscope were used to determine the sensitivity of GCV,5-FC or both on prostate cancer cells(PC-3m)after being transfected with CD-TK gene.Results:Significant cytotoxic activities were found by GCV or5-FC on CD-TK gene-trans-duced prostate cancer cells,and simultaneous treatment with two prodrugs resulted in synergistic toxici-ty.Conclusion:The experiment indicated that CD-TK gene therapy has significant antitumor activities on transfected PC-3m cells in vitro,superior to single suicide gene therapy.
