细胞因子裸质粒DNA的抗肿瘤研究

摘要 目的 探讨瘤体内直接注射细胞因子裸质粒DNA (pcDNA3.1-GM -CSF和 pcDNA 3.0 -IL -15基因 )对小鼠肺腺癌的治疗作用。方法 将 pcDNA 3.1-GM -CSF基因和 pcDNA 3.0 -IL -15基因 2种细胞因子基因直接注射到小鼠肺腺癌瘤体内 ,采用病理分析和免疫组化方法 ,对 pcDNA 3.1-GM -CSF和pcDNA 3.0 -IL -15转基因治疗后的小鼠肿瘤组织进行分析。结果 应用 pcDNA3.1-GM -CSF和 pcDNA 3.0 -IL -15裸质粒DNA ,通过直接注射均可抑制肿瘤生长。经pcDNA 3.1-GM-CSF和 pcDNA 3.0 -IL -15基因治疗后 ,肺癌组织内可见有部分坏死 ,瘤体内有大量炎性细胞及部分树突状细胞浸润。细胞因子GM -CSF和pcDNA 3.0 -IL -15基因在肿瘤组织中有不同程度的表达。结论 细胞因子裸质粒pcDNA 3.1-GM -CSF和pcDNA3.0 -IL -15直接注射 ,可以在组织中表达 。 Objective To investigate the therapeutic effect against mouse pulmonary adenocarcinoma by injection with naked plasmid DNA encoding granulocyte-macrophage colony stimulating factor(GM-CSF) and Interleukin-15(IL-15) into tumor.Methods Plasmids of pcDNA3.1-GM-CSF gene and pcDNA3.0-IL-15 gene were injected into mouse pulmonary andenocarcinoma.The mouse tumor tissue was analyzed by pathological analysis and immunohistochemistry after transfected gene therapy with GM-SCF and IL-15.Results Both GM-CSF and IL-15 plasmid DNA inhibited the growth of tumor.Part of pulmonary adenocarcinoma tissue was necrotic and a large number of infiltrating immune cells and some invasive dendritic cells could be observed in tumor after gene therapy with GM-CSF and IL-15.Both GM-CSF and IL-15 gene could express in tumor tissue at different level.Conclusion Plasmid DNA encoding GM-CSF and IL-15 can express in tumor tissue by injection into tumor and have a significant anti-tumor effect.
