血管内皮生长因子反义cDNA抗血管生成作用的体外实验研究

摘要 目的 :研究血管内皮生长因子 (VEGF)反义cDNA对血管内皮细胞VEGFmRNA和蛋白表达的影响 ,并探讨其对内皮细胞生长及增殖过程的作用。方法 :以脂质体介导VEGF反义cDNA转染血管内皮细胞 ,通过原位杂交法检测细胞VEGFmRNA、免疫组化法检测细胞VEGF蛋白和PCNA增殖活性、ELISA法检测分泌型VEGF蛋白、MTT法测定细胞生长率 ,研究VEGF反义cDNA对内皮细胞的作用。结果 :转染后血管内皮细胞内源性VEGFmRNA和蛋白的表达、细胞生长及增殖受抑。结论 Objective:To study the effects of VEGF_antisense_cDNA to vascular endothelial cells on the proˉduction of VEGF and the process of cell growth and proliferation.Methods:Vascular endothelial cells were transfected with VEGF_antisense_cDNA via lipofectamine.We investigated the production of VEGF and the process of cell growth and proliferation by hybridyzation in situ,immunohistochemistry,ELISA,and MTT.Reˉsults:After transfection,the expression of intrinsic VEGF mRNA and protein in vascular endothelial cells deˉcreased,and cell growth and proliferation were inhibited.Conclusion:Transfection of VEGF_antisence_cDNA can down_regulate the production of VEGF and inhibit the process of cell growth and proliferation.
