摘要
目的 :探讨人参总甙、人参皂甙单体对脐血CD34 +细胞体外增殖及分化的影响。方法 :用MethoCultTMH4435半固体培养基加入不同浓度的人参总皂甙 (GS)、Rg1、Rb1,计数CFU -E、BFU -E、CFU -GM集落。结果 :GS、人参皂甙单体Rg1、Rb1具有刺激脐血CD34+ 细胞集落形成的作用 ,人参皂甙单体的作用优于GS。不同浓度的人参皂甙对脐血CD34+ 细胞集落刺激不同 ,但其集落形成能力明显高于对照组。GS的最佳浓度为50mg/L,Rg1和Rb1最佳浓度为5mg/L。高浓度的GS、Rg1、Rb1集落形成能力下降 ,但并不显示抑制作用。结论 :人参总甙、人参皂甙单体能促进造血干或祖细胞增殖 ,并且能诱导定向分化 。
Objective:To investigate the effects of ginsenosides,Rg1and Rb1on the proliferation and difˉferentiation of CD34 + cell isolated from cord blood in vitro.Methods:CD34 + hematopoietic stem cell was isolated and purified from human cord blood.The cells were placed in semisolid medium containing GS,Rg1,and Rb1with different concentrations.The numbers of colonies were counted on7or14days respectively.Results:The numbers of GS,Rg1,Rb1,CFU_E,BFU_E,and CFU_GM were higher than that in control group.The effects of Rg1and Rb1were superior to GS.The optimal concentration of GS was50mg/L,Rg1was5mg/L,and Rb1was5mg/L.Conclusion:GS,Rg1,and Rb1could stimulate the proliferation and differentiation of the hematopoietic stem cell,and ginsenosides had analogous and synergetic effects with the cytokines.
