淹水胁迫下不同耐渍性玉米自交系根系中的酶学研究 被引量:13

摘要 以 4种耐渍性存在差异的玉米自交系为材料 ,对淹水前后玉米根系中乙醇脱氢酶 (ADH)、NAD+-苹果酸酶 (ME)、谷氨酸脱氢酶 (GDH)和过氧化物酶 (POD)的表达量及其同工酶的变化进行研究 ,结果表明 :淹水 3d后的各个自交系根系中 ,ADH的活性都明显增加 ,Adh2位点被诱导表达 ;耐渍性较差的NC2 86和黄早四根系中ME活性分别增加 170 %和 130 %以上 ,耐渍性较好的自交系ME活性则变化不明显 ;除NC2 86外 ,GDH活性在淹水前后变化不明显 ,但淹水处理后有一同工酶位点Gdh4被诱导表达 ;淹水后 ,凤可 1根系POD活性略有上升 ,其他材料POD活性均有不同程度下降。作者认为 ,玉米根系在缺氧条件下快速进入乙醇发酵途径和氮代谢等途径 ,提供生存所需能量和减少代谢有毒中间产物积累 ,是玉米耐渍性的基本特征。 Four maize inbreds which display differentially endurance to waterlogging were chosen to assay the expression and allozyme gel pattern of four enzymes before and after waterlogging in maize root.Alcohol dehydrogenase(ADH),NAD + -Malic enzyme(ME),Glutamate dehydrogenase (GDH) and Peroxidase (POD)were assaied.The result showed that activity of ADH was increased distinctly and expression of adh2 loci was induced by waterlogging stress in 3 days late.Activity of ME increased more than 170% and 130% respectively in root of both lines,NC286 and Huangzaosi,which were weakly tolerance to flooding,but didn't vary non-significantly in strongly tolerance inbred.Activity of GDH had little change in studied inbred except NC286,but the expression of a new allozymic loci involve in gdh4 was induced after waterlogging 3 days.Activity of POD was decreased slightly in maize root except Fengke 1.We assume that a mechanism quickly transfer to alcoholic fermentation and nitrogen metabolic pathway could supply essential energy,and discompose poisonous metabolic products for maize survive when root be grow in oxygen deficiency environment,the responses would be basic characteristics of maize tolerance to waterlogging.
