摘要
分离纯化的黄瓜子叶原生质体 ,以 5 5× 10~ 5 5× 10 5个 /mL的不同密度培养于mKM8p液体培养基中 ,均获得不同程度的细胞分裂。当培养密度超过 1 0× 10 3 个 /mL时 ,原生质体可持续分裂至愈伤组织形成 ,最高植板率为 5 4%。原生质体来源的愈伤组织经诱导培养产生大量体胚并再生成植株。Ca2 + 浓度对黄瓜原生质体的稳定和细胞分裂有重要影响。
Isolated and purified cotyledon protoplasts of cucumber (Cucummis sativus L.'Cuixiu')were cultured at different population density ranging from 5.5×10/mL to 5.5×10 5/mL in mKM8p liquid medium respectively and underwent cell division in varying degrees.Sustained cell divisions with eventual somatic colony production occurred only when the population density of protoplasts exceeded 1.0×10 3/mL,the highest plating efficiency was about 54%.Large quantities of embryoids differentiated from protoplast derived calli and then converted into plantlets after succedent induction culture.Results demonstrate that the concentration of Ca 2+ has an important effect on the stability and cell division of cucumber protoplasts.
