恒磁场对人脐静脉内皮细胞增殖的影响 实验证据支持磁场效应可降低RS发生率 被引量:7

摘要 目的观察恒磁场(staticmagneticfield,SMF)对人脐静脉内皮细胞(humanumbilicalveinendotheliumcell,HUVEC)增殖的影响,以探讨SMF能否用于经皮腔内冠状动脉成形术(percutaneoustrans-luminalcoronaryangioplasty,PTCA)及支架植入术后冠脉再狭窄(restenosis,RS)的防治。方法用含100g/L小牛血清的DMEM培养液体外培养人脐静脉内皮细胞(ECV304),分别在给予SMF和无SMF条件下培养,用3H-胸腺嘧啶脱氧核苷(3H-TdR)掺入法和流式细胞技术分析SMF对HUMEC的DNA合成及细胞周期的影响。结果对照组cpm值为1194.669±287.250,恒磁场组cpm值为1356.578±363.845,差异显著(P<0.05)。恒磁场组S期细胞高于对照组,G1期细胞较对照组降低,增殖指数PI值(S+G2M)%增高,差异显著(P<0.05)。结论SMF对HUMEC的增殖具有明显的促进作用。SMF对PTCA支架植入术后的冠脉RS可能具有防治作用。 Aim To investigate the adjusting effect of static magnetic field (SMF) on the proliferation of human umbilical vein endothelial cells(HUVECs), and observe if SMF can be used to treat the coronary restenosis after percutaneous transluminal coronary angioplasty(PTCA) and in stent. Methods Human umbilical vein endothelial cells (ECV304) were cultured in vitro with DMEM and 10 percent calf plasma.The HUVECs were cultured with SMF and without SMF respectively.The amount of HUVECs proliferation was monitored by 3H TdR incorporation and flow cytometric(FCM) analysis.Results The cpm of control group was 1 194.669± 287.250,which SMF group was 1 356.578± 363.845( P< 0.05) .The cell cycle tested through FCM method showed that cells of the S phase in the SMF group were higher than those of the control group, cells of the G1 phase in the SMF group were lower than those of the control group, the proliferation index(PI)were increased markedly in the SMF group( P< 0.05) . Conclusion The HUVECs proliferation is significantly promoted by SMF.The results suggest that possibility of using magnetic field to prevent and cure the coronary restenosis after percutaneous transluminal coronary angioplasty(PTCA) and in stent.<P>
