摘要
用短时间超速离心结合制备性 SDS 聚丙烯酰胺凝胶电泳纯化人血浆 apoE.以此纯品免疫家兔制备了单价兔抗人 apoE 抗血清,与常规方法相比,本法具有操作简单、费用低和提纯周期短等优点,是一种简单实用的方法。
This article presents a two steps procedure of apoE purification whith included a short-time ultra- centrifugation and preparative sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS- PAGE).The molecular weight and immunological properties of isolated apoE Were verified by poly- acrylamide gel electrophoresis in SDS and urea.The monospecific rabbit anti-human apoE antiserum. prepared by immunizing New Zealand white rabbits,was verified by double diffusion and immuno- electrophoresis.In contrast to the common method of apoE isolation,the present method is rapid,eco- nomical,needs no special apparatus and can be used as a suitable method for apoE isolation.
