用序列超速离心法从血清中分离高密度脂蛋白(HDL),经0.5％琼脂糖凝胶电泳显示为一条区带。HDL载脂蛋白(apoHDL)先经Sephadex G—200凝胶过滤柱层析,取峰Ⅱ中部洗脱液,再通过DE—52离子交换柱层析分离出载脂蛋白A—Ⅰ(apoA—Ⅰ),用0.1％SDS—PAGE证实为单一区带,分子量为28000道尔顿。
Human serum high density lipoprotein was isolated by a process of sequence ultracentrifugation, yielding a single band on 0.5% agarose gel electrophoresis and contained no albumin. The highdensity lipoprotein was delipidated and fractionated by sephadexG—200 chromatography into four peaks. The 2nd peak was taken and isolated by DEAE—52 ionexchange chromatography. The apolipoprotein A—I was shown to be pure, which yielded a single band with a molecular weight 28000 when analyzing with 0.1% SDS—10% polyacylamide gel electrophoresis.
