摘要
以发根农杆菌转化苦豆子的子叶和下胚轴外植体,结果表明,辅助以超声波处理有助于转化率的提高,当超声波(功率120W,震荡频率50kHz)处理25min时,转化率达到最高峰(子叶为83.7%,下胚轴为39.1%),分别高于对照(14.4%和9.8%)69.3%和29.3%。在此条件下,进一步添加AS100μmol/L,可使转化率再进一步提高9.9%和7.6%。
The effect of additional ultrasonic treatment on transformating rate of Sophora alopecuroides me-diated by Agrobacterium rhizogenes was studied in the present paper. The highest rates,83. 7% for cytole-don and 39. 1% for hypocotyl explants,were achieved after 25 min of treatment (120 W,50 kHz),being 69. 3% and 29. 3% higher than control (14. 4% and 9. 8%). Meanwhile,the supplement of 100 μmol/L acetosyringone (AS) to A. rhizogenes suspension could further increase the rate by 9. 9% and 7. 6% for cy-toledon and hypocotyl explants,respectively.
