摘要
目的 研究全脑缺血再灌注后大鼠脑神经细胞的凋亡情况。方法 SpragueDawley大鼠 2 8只 ,雌雄各半 ,随机分 5组 ,即对照组 (假手术组 )以及复苏后 3、6、12、2 4h组 ,对照组 4只 ,其余每组 6只。采用窒息合并冰氯化钾停跳液致大鼠心跳骤停—心肺复苏模型 ,停跳 5min后开始心肺复苏 ,采用TUNEL方法和透射电镜观察复苏后神经细胞的凋亡情况。结果 复苏后 3h大鼠脑皮质神经元就出现细胞凋亡 ,复苏后 2 4h海马区TUNEL阳性率为 6 0 %。
Objective To investigate the apoptosis of neuronal cell after global ischemia-reperfusion.Methods Cardiac arrest was induced by asphyxiation and ice-cold 0.5 M KCl in rats and resuscitation efforts would begin five munites after arrest. 28 Sprague Dawley rats, half female and half male, were randomly divided into 5 groups: sham control group (n=4); after cardiopulmonary resuscitation rats were allowed to reperfuse spontaneously for 3, 6, 12 and 24 hours(n=6,each group). The temporospatial cellular reactions were assessed by terminal deoxynucleotidyltransferase-mediated dutp-FITC nick endlabeling (TUNEL) and transmission electron microscope (TEM). Results TEM revealed early onset apoptosis in cortex 3 hours after resuscitation. At 12 hours post-resuscitation, TUNEL-positive cells could be observed in the hippocampal. There were 6.0% TUNEL-positive cells at 24 hours. Conclusion Apoptosis was one mechanism of neuronal cell damage after global ischemia-reperfusion.
