白眉蝮蛇蛇毒精氨酸酯酶致畸实验 被引量:1

TERATOGENICITY TEST OF ARGININE ESTERASE FROM AGKISTRODON HALYS USSURIENSIS VENOM

摘要 目的 :研究白眉蝮蛇(Agkistrodonhalysussuriensis)蛇毒精氨酸酯酶对大鼠的致畸胎作用 ,为临床应用提供依据。 方法 :用大鼠进行致畸胎试验 ,将动物分成5组 :阴性对照组(生理盐水)、精氨酸酯酶低剂量组(0.06U/kg)、中剂量组(0.18U/kg)、高剂量组(0.36U/kg)和阳性对照组(VitAD滴液10mg/kg)。精氨酸酯酶于受精第6d连续静脉给药至15d,第20d(分娩前1d)断头处死动物并剖腹。通过3项指标确定精氨酸酯酶的致畸胎作用 :一般综合指标 ,包括实验期间动物的体重、进食量、精神状态和活动情况 ;妊娠指标 ,包括剖腹检查记录的着床总数、死胎数、活胎数、吸收胎仔数、胚胎体重、身长、尾长和性别比 ;致畸指标 ,包括对外形、器官和骨骼畸胎率的测定。 结果 :母鼠胚胎毒性症状出现在高剂量组(0.36U/kg) ,与对照组相比表现为妊娠期间平均体重增长减慢和胎仔的体重减轻(P<0.05) ,但外形、内脏和骨骼畸形率与对照组相比未见明显差异(P>0.05)。其它药物处理组与对照组相比没有明显差别(P>0.05)。 结论 :纯精氨酸酯酶0.18U/kg以下对孕鼠无胚胎毒性和致畸作用 ,高于0.18U/kg则有一定的胚胎毒性。临床应用的剂量(0.012U/kg) Purpose: To study the teratogenic effect of arginin e esterase from Agkistrodon halys ussuriensis venom for the cl inical application of arginine esterase in the future. Methods: SD rats were divided into five groups: negative control grou p (normal saline), arginine esterase low dose group (0.06 U / kg), middle dose group (0.18 U / kg), high dose group (0.36 U / kg) and positive control group (vitamine AD). The arginine esterase was given by i.v. continuously for 10 days from th e 6th day of pregnancy to the 15th day. Rats were sacrific ed one day before birth. The teratogenic effect was determined by evaluating and comparing general criteria (including locomot or activity, growth rate, appetite and death rate), cyesis cri teria (including implantation number, died fetus number, living fetus number, resorption embryo number, fetus body weight, fetus body length, fetus tail length and fetus sex ratio), and teratosis criteria (including external appearance teratosis rate, internal organ teratosis rate and skeleton teratosis rate). Resu lts: The body weight of the female rats and the fetuses de creased at the dosage of 0.36 U / kg compared with the negat ive control animals (P<0.05). But there were no significant di fferences in teratosis rates(P>0.05). This result showed that ar ginine esterase had toxic effect on fetus but had no teratog enic effect even at high dosage. There were no significant d ifferences in other criteria between the lower dose arginine e sterase treated animals and the negative controls(P>0.05). Conclos ion: The safe dosage of arginine esterase for rats in terato genic effect is 0.18 U / kg and it has toxic effect to emb ryo at higher dosage. The dosage of 0.18 U / kg is eq uivalent to 15 times of the clinical dosage of 'Qingshuanmei' whose main component is arginine esterase, and it indicated that clinical usage of the arginine esterase is safe.
