摘要
比较MTT比色分析法、中性红摄取法、结晶紫染色湿测法、结晶紫染色干测法和~3H-TdR摄入法五种方法检测L929细胞毒活性的敏感性和稳定性。结果表明五种方法检测的TNF活性有明显差异,以结晶紫染色干测法具有较敏感、稳定、简便之优点.并用此方法检测TNF对三种不同肿瘤细胞的细胞毒活性结果,不同的肿瘤细胞对TNF的敏感性有明显差异,提示可用结晶紫染色干测法检测TNF对不同肿瘤细胞的细胞毒性效应的敏感性。以供临床治疗参考。
The comparative study of the methods of MTT, neutral red uptake, crystal violet wet assay, crystal violet dry assay and ~3H-TdR incorporation was done to assay the cytotoxic sensitivity and stability of the tumor necrosis factor (TNF). The results showed that there was significant difference among the five methods, of which the crystal violet dry assay bore the advantages of higher sensitivity, stability, and simplicity than the others did. By assaying the cytotoxic effect of the three types of tumor cells, it was found that there was significant difference in the sensitivity of different tumor cells to TNF. Thus, the method of crystal violet assay can be used to test the sensitivity of TNF to the cytotoxic effect of tumor cells.
