摘要
目的 建立定量检测脑脊液 (CSF)结核分枝杆菌聚合酶链反应 (PCR)分子灯塔法并评估其临床应用价值。方法 应用PCR分子灯塔定量技术检测 83例CSF结核分枝杆菌并与抗酸杆菌染色、结核抗体酶联免疫吸附试验 (结核抗体ELISA ,简称酶联OT)进行比较。结果 PCR分子灯塔法能准确、定量地检测脑脊液结核分枝杆菌 ,其阳性检出率 ( 88.3 %)分别高于酶联OT( 2 3 .2 %)、抗酸杆菌染色 ( 0 %)的检出率。结论 PCR分子灯塔法用于定量检测CSF结核分枝杆菌具有简便、快速、灵敏度高、特异性强等优点。在监测患者病情、预后、指导用药方面有着广泛的应用前景 ,值得在临床上推广应用。
Objective To develop a quantitative method for detection of Mycobacterium tuberculosis in cerebrospingal fluid (CSF) and evaluate its clinical value.Methods The CSF specimens of 83 patients were tested by smears, Enzyme-linked immunosorbent assays (ELISA) and Polymerase chain reaction (PCR) molecular beacon assays.Results The sensitivities of PCR-molecular beacon assay, ELISA and smear in detecting Mycobacterium tuberculosis of CSF speimens of tuberculous meningitis patients were 88.3%, 23.2% and 0%, respectively.Conclusions The PCR-molecular beacon assay is a simple, rapid, specific and sensitive way for detecting Mycobacterium tuberculosis in CSF.It is also and useful tool for monitoring patient's condition and conducting the treatments.
