河豚毒素单用及与吗啡合用对大鼠福尔马林致痛的影响 被引量:4

摘要 目的:探讨河豚毒素(tetrodoxin,TTX)单用及与吗啡合用的镇痛效应。方法:采用大鼠福尔马林致痛实验,分别单独肌注河豚毒素及将微量河豚毒素(0.39μg·kg^(-1),0.19μg·kg^(-1))与吗啡联合给药,比较合用是否具有镇痛增强或协同效应。采用概率单位法计算镇痛的半数抑制量(ID_(50))。结果:TTX的镇痛半数抑制量(ID_(50))为0.8μg·kg^(-1);单用吗啡的ID_(50)为2.6mg·kg^(-1);TTX0.39μg·kg^(-1)(1/50 LD_(50))和0.19μs·kg^(-1)(1/100 LD_(50))与吗啡合用,吗啡的镇痛ID_(50)分别下降为0.5mg·kg^(-1),1.1mg·kg^(-1),联合用药的性质经检验具有协同作用。结论:河豚毒素对福尔马林所致疼痛具有镇痛作用,与吗啡联合应用具有协同镇痛效应。 ObjectiveTo explore the effects of tetrodotoxin (TTX) alone and combined with morphine on formalin-induced pain test in rats. Methods Formalin test in rats was used for nociceptive test. TTX, morphine, or both were administered i. m. to rats and the analgesic effects were measured. Data were expressed as the median inhibitory dose( ID50). Results The ID50 of TTX alone was 0. 8μg · kg-1. The ID50of Morphine alone was 2. 6 mg · kg-1. The combination of TTX (0. 39μg · kg-1 or 0. 19μg · kg-1) and morphine showed more potent than each of them alone. The ID50 of Morphine reduced to 0. 5mg · kg-1 ,.1. 1mg · kg-1, respectively. An isobologram showed the synergistic effect between TTX and morphine. Conclution The result indicated that TTX had analgesic effect in the formalin test in rats. Comparing the effects of TTX alone and combined with morphine, the latter revealed a synergistic effect.
