摘要
介绍了^(90)Y标记DTPA-抗人肝癌铁蛋白抗体联结物的标记方法及标记物的分离和分析方法,并对几种标记方法进行了比较,认为用微量离心柱分离DTPA-抗体联结物,而后进行^(90)Y的标记,效果最好,标记率高达92%。标记物经进一步纯化,其放化纯度可达99%以上。抗体经标记后免疫活性无明显损失。实验结果还表明,DTPA-抗体联结物和^(90)Y的稀释效应及标记体系中杂质金属离子的存在均对^(90)Y的标记有极大的影响。
This paper presents the method of labelling DTPA - Ab (antiferretin antibody of human hepatocellular carcinoma )conjugate with 90Y as well as the analytical method of the labelled compounds. The labelling percentage of the DTPA - Ab conjugate separated by microcen-trifuged column gel filtration using Sephadex G -75 is as high as 92% . After further purification, the radiochemical purity can be higher than 99%. The experimental results also show that the dilution factor of DTPA - Ab conjugate and 90Y as well as the presence of impurity metal ions have great influence on the 90Y labelling of DTPA - Ab conjugate.
