摘要
本实验观察了冷冻损伤后大鼠血清中血管紧张素Ⅰ转换酶(ACE)活性的改变及其与冷冻损伤程序度的关系。150只Wistar大鼠乙醚麻醉后双后足在-25℃乙醇中浸冻至组织温度分别达到-20℃、-10℃和0℃,冻后4小时、24小时、3天、5天和7天用紫外分光光度法测定血清中ACE活性。结果表明,冻后大鼠血清中ACE活性呈不同程度的增加,其增加程度与冷冻损伤程度有关。提示冷冻损伤可引起不同程度的血管内皮细胞损伤,使之释放到血流中的ACE活性增加。
It has been known that angiotensin I converting enzyme (ACE)from endothelial cells acts as an important factor in converting plasma angiotensin Ⅰ into the powerful vasoconstrictor angiotensin Ⅱ.The present experimental study was performed to observe the changes of ACE activity in serum of rats after freezing,and the relationship of these changes with degrees of cold injury. A total of 150 Wistar rats weighing 250+20g were used . Under ether anesthesia, the rat's hind feet were immersed in a freezing solution of alcohol(-25°C)until the temperature of tissue reached - 20°C, -10°C,and 0°C respectively in order to produce different degrees of frostbite. The serum ACE activity were determined by means of ultraviolet-spectrophotometry (wavelength 228nm). Serum ACE activity in the normal control rats(n=15) was 46.55+2.36 nmol/ml. min(M+S.E.);in-20°C rats, ACE activnties were 60.9卤4.3, 72.5+2.9, 66.8+3.4, 63.8+4.4 and 59.2+5.2 respectively at 4h,l,3,5 and 7 days after freezing;in -10°C group they were 59.6+3.8, 59.5+5.6, 51.5卤2.8, 55.6卤4.2 and 45.0 + 2.3 respectively; in 0°C group they were 49.0卤3.9, 56.2+2.1, 49.6卤2.6,43.5+4.0,and 47.6+3.9 respectively (each is mean of nine aminals). The results showed that the increases in serum ACE activity were closely related to the degrees of cold injury. It suggests that the freezing damage of vascular endothelial cells may lead to an increase in the release of ACE, which may play an important role in the pathological process in cold injury.
