褪黑素对卡因酸介导大鼠海马神经元毒性的影响 被引量:1

摘要 目的 探讨褪黑素对腹腔注射卡因酸介导大鼠海马神经元损害的影响。方法 将大鼠随机分成 3组 ,实验组给予腹腔注射卡因酸 ,治疗组给予腹腔注射卡因酸和褪黑素 ,对照组给予腹腔注射等体积磷酸缓冲液 (PBS) ;3天后以尼氏染色法检测大鼠海马神经元 ,DNA缺口末端标记 (TUNEL)法检测海马神经元凋亡。结果 大鼠海马神经元计数 ,尼氏染色实验组及治疗组明显少于对照组 (P <0 .0 1 ) ,治疗组明显高于实验组 (P<0 .0 1 ) ;TUNEL染色阳性细胞数治疗组明显低于实验组 (P <0 .0 1 )。 Objective To study effect of melatonin on the hippocampal neuron apoptosis of rats by kainite. Methods The rats were divided into three groups randomly: the experimental group was made with the intraperitoned administration of kainite, the KA+MT group was made with the kainite and melatonin,the control group was made with phosphate buffer solution (PBS) in some way. After 3 days,the lose of neuron in hippocampus of rats was detected by Nissl staining, The neuronic apoptosis in the hippocampus were examined with terminal deoxynucleotidyl transferase mediated d UTP biotin nick end labeling (TUNEL).Results The neurons of in the hippocampus of experimental and KA+MT rats were significantly decreased than that of control rats (P<0.01), KA+MT rats were significantly increased than that of experimental rats (P<0.01).The TUNEL positive cells of in the hippocampus of experimental rats were significantly increased than that of KA+MT rats (P<0.01). Conclusion The result of this study indicated that melatonin can obviously inhibit the neuronic apoptosis of kainite in brain of rat.
