摘要
目的 建立一种血清锰的石墨炉原子吸收分光光度分析方法。方法 将血清和 0 44mol/L的HNO3按 1 9稀释后 ,1 5 0 0 0r/min高速离心 1 0min ,取上清 2 0 μl直接测定。结果 发现灰化温度为 1 30 0℃ ,原子化温度为1 90 0℃ ,净化温度为 2 4 50℃和基底改进剂为 5 μg钯 +3μg硝酸镁最佳 ;平均回收率可达到 99 56 % ;变异系数 (CV)为5 0 % ;特征浓度为 6 376 1 0 pg /0 0 0 4 4A ,检出限为 6 7pg。结论 本方法为一种可信度较高。
Objective To set up a new method of detecting the l evel of manganese in serum with the graphite atomic absorption spectroscopy(AAS) . Methods The serum was diluted with HNO 3 and centrifuged 10 minu tes by 15 000 r/min first and the solution above in the tube was going to be determined. Results The optimal temperatures for cineration, atomi zation and clearance were 1 300 ℃, 1 900 ℃ and 2 450 ℃ with the matri x modifier of a unit of 5 μg Pd and 3 μg Mg(NO 3) 2, respectively. The avera ge reclamation rate was 99 56% and the coefficient of variation was about 5%; t he sensitivity was 6 376 10 pg / 0 004 4 A and the minimal level could be detected was 6 7 pg. Conclusion This is a practicable and cre dible method for detection serum Mn with AAS.
