摘要
目的 探讨辐射对培养小鼠骨髓基质细胞表达血管内皮生长因子 (VEGF)的影响及凋亡率变化。方法 采用流式细胞术和 RT- PCR法。结果 经 8.0 Gy射线照射后 1h,骨髓基质细胞 VEGF m RNA水平即较正常对照显著升高 ,12 h达到高峰 ,2 4 h恢复至正常水平 ;照射后不同时间 ,骨髓基质细胞凋亡率与正常对照无显著差异。结论 8.0Gy6 0 Co- γ射线照射后骨髓基质细胞中 VEGF m RNA表达增强 。
Objective To explore the changes of vascular endothelial growth factor (VEGF) expression and apoptosis in cultured bone marrow stromal cells (BMSCs) after exposure to γ radiation. Methods RT PCR and FACs were used. Results The expression level of VEGF mRNA in BMSCs was elevated after exposure to 60 Co γ rays at the dose of 8 0 Gy. The expression peak was at 12 h after irradiation. The apoptosis rate of BMSCs had no significant changes. Conclusion The elevation of VEGF expression in BMSCs by irradiation might be involved in the protection of BMSCs and in the recovery of hematopoiesis after γ irradiation.
