摘要
目的 :探讨广西肥胖症、非胰岛素依赖性糖尿病 (NIDDM)是否与肥胖基因突变有关 ,同时观察广西壮族汉族人群的肥胖基因 (ob基因 )多态性。方法 :将 2 96例研究对象分为三组 :肥胖 4 7例 ,NIDDM79例 ,对照组 170例 ,用 PCR法扩增各组 ob基因的第一外显子 ,再用改良的 SSCP法对 PCR扩增片段进行分析比较 ,并对异常扩增片段测序。结果 :广西壮族人 ob基因第一外显子存在多态性 A75→ T。结论 :ob基因第一外显子突变并非导致肥胖症。
Objective:To study the relation between obesity, NIDDM and ob gene mutation, and to observe the ob gene polymorphism of Zhuang and Han peoples in Guangxi.Methods:296 cases were divided into three groups (obesity 47, NIDDM 79, control 170), PCR SSCP was used to clone and analyze the first exon of their ob gene, and the abnormal ob gene fragments were sequenced to check their mutation.Result:Polymorphism(A75→T) was observed in the first exon of ob gene in Zhuang people from Guangxi. Conclusion:The mutation in the first exon of ob gene is not the common reason that results in obesity and NIDDM.
