肺癌患者血浆游离性肿瘤DNA的研究

摘要 目的 研究肺癌患者血浆游离DNA微卫星不稳定和杂合性缺失在肺癌诊断中的价值。方法 通过PCR 银染法检测肺癌病人的 37例新鲜手术标本、94例全血标本DNA 3p部位三个微卫星位点的LOH、MSI。结果 大多数肺癌患者的血浆游离DNA的浓度在微克水平以上。联合三个位点进行LOH、MSI检测时 ,组织和血浆的阳性率均在 70 %以上。不同分期、不同病理类型之间差异不显著 ;有无淋巴结转移之间差异有显著性意义。结论 肺癌患者血浆游离DNA可以作为基因检测的新途径。 Objective To study the diagnostic value of LOH and MSI of 3p microsatellite point in free plasma DNA of lung cancer patients. Methods We use techniques of PCR and sliver dye to detect LOH and MSI of three microsatellite sites of plasma DNA on chromosome 3p from 37 surgery fresh tissues and whole blood sample from 94 patients. Results The plasma free DNA concentration of the most lung cancer patients was above microgramme level. Above 70% of tissues and plasmas were positive. No significant difference was found on different stages and pathology types. The result of patients with lymph node metastasis was significant different with the patients without lymph node metastasis. Conclusion Plasma DNA of lung cancer is a good medium of gene detection and will be used widespread in the future.\;
