摘要
目的 :观察当归补血汤血清对SMMC 772 1增殖影响的量效和时效关系 .方法 :采用纯种新西兰雄兔 2 5只 ,随机分 5组 ,分别给予 5 0mL的 32 ,16 ,8和 4g·kg-1当归补血汤和生理盐水灌胃 ,于不同时间点收集血清 ,以MTT法测定上述血清对人肝癌细胞系SMMC 772 1增殖抑制作用 .结果 :当归补血汤血清对人肝癌细胞SMMC 772 1增殖有抑制作用 ,且具有时间和剂量依赖性 ,灌胃后 4 5min抑制作用最强 ,各组抑制强度分别为 0 .4 6± 0 .0 5 (32g·kg-1,P <0 .0 1) ,0 .5 3± 0 .0 6(16g·kg-1,P <0 .0 1) ,0 .5 5± 0 .0 4 (8g·kg-1,P <0 .0 5 ) ,0 .6 1± 0 .0 3(4g·kg-1,P >0 .0 5 ) .ED5 0为 12 2g·kg-1;效应半衰期为 2 .2 0h .结论 :当归补血汤经肠道进入体内具有一定的抑制肝癌细胞增殖的作用 。
AIM: To observe the effects of the serum of DangGuiBuXue Decoction (DGBXD) on the proliferation of SMMC 7721. METHODS: 25 rabbits were divided into 5 groups at random and the rabbits sera of different time points were collected after ig 50 mL of Naltrii chlorcid (9 g·L -1 ) or DGBXD with the dosage of 32 g, 16, 8, 4 g·kg -1 . The culture medium of human heptocarinoma cell lines of SMMC 7721 was treated by the rabbits sera. Effects of the sera on inhabiting proliferation of SMMC 7721 were detected by MTT assay. RESULTS: The sera of DGBXD ig inhibited the proliferation of SMMC 7721 dose and time dependent manner. Tmax was 45min after ig and the maximum effects of BGBXD was 0.46±0.05 (32 g·kg -1 , P <0.01), 0.53±0.06 (16 g·kg -1 , P <0.01), 0.55±0.04 (8 g·kg -1 , P <0.05), 0.61±0.03 (4 g·kg -1 , P >0.05) vs control. ED50 was 122 g·kg -1 and T 1/2 was 2.2 h. CONCLUSION: The sera of DangGuiBuXue Decoction can inhibit the proliferation of human liver cancer SMMC 7721 in a dose and time dependent manner.
