神经生长因子对N-甲基-D-门冬氨酸诱导人胚胎RPE细胞凋亡的保护作用 被引量:1

摘要 目的 探讨神经生长因子(NGF)对体外培养的人胚胎视网膜色素上皮(hFRPE)细胞凋亡的保护作用。方法 将传代培养的hFRPE细胞,用200μmol/L,300μmol/L N-甲基-D-门冬氨酸(NMDA)建立诱导的hFRPE细胞凋亡模型,并用吖啶橙(AO)荧光染色计数和透射电镜(TEM)观察NGF对凋亡细胞的保护作用。 结果 用AO荧光染色及TEM均观察到经200 μmol/L,300μmol/L NMDA处理72 h后的hFRPE细胞出现典型的凋亡形态学改变。200μmol/L.NMDA作用72 h后hFRPE细胞凋亡指数为(32.587 9±1.488 0)%和200 μmol/L NMDA+300 μmg/L NGF作用72 h后的hFRPE细胞凋亡指数(20.768 7±1.302 4)%相比,差异有显著性(q=2.941 4,P=0.005 0);300 μmol/L NMDA作用72 h后hFRPE细胞凋亡指数为(42.506 4±3.014 8)%,与300 μmol/L NMDA+300 μg/L NGF作用72 h后的hFRPE细胞凋亡指数(25.687 2±1.8974)%相比,差异有非常显著性的意义(q=4.117 8,P=0.000 0)。 结论 NCF能拈抗NMDA诱导的hFRPE细胞凋亡。 Objective To investigate the protective effect of nerve growth factor ( NGF) on apoptosis of cultured human fetal retinal pigment epithelial cells ( hFRPE) induced by N-methyl-D-aspartate (NMDA) in vitro. Methods Subeultured hFRPE cells were treated with different concentrations of NMDA to establish apoptotic model. The protective effect of NGF on apoptosis of cultured hRPE cells were assessed using an acridine orange ( AO) staining method and transmission electron microscopy ( TEM ). Results hFRPE were exposed to NMDA 200 μmol/L, 300 μmol/L for 72 hs and typical apoptosis morphological changes included condensed chromation .nuclear fragmentation and reduction of nuclear size and cell volume. There was a statistical difference in hFRPE cells with apoptosis between 200 μmol/L NMDA +300μg/L NGF and 200μmol/L NMDA groups (q-tesl,q =2. 941 4,P = 0. 005 0) ,in 300 μmol/L NMDA +300 μg/L NGF and 300 μmol/L NMDA as well (q-test,q = 4. 117 8 ,P =0. 000 0). Conclusion NGF has a protective effect on NMDA-induced hFRPE cells apoptosis.
