摘要
目的 :研究重组干扰素 -γ对哮喘患者外周血单核细胞 (PBMC)IL - 4和IL - 10mRNA表达的调节作用。方法 :哮喘患者9例、正常人 5例的外周静脉血 ,ficoll-hypaue密度梯度离心分离PBMC ,分为 2组进行培养 ,A组加入rIFN -γ ,0 .2U·μl-1;B组为不加rIFN -γ的对照组。在 3 7℃ ,5 %CO2 培养箱中培养 48h后 ,用RT -PCR方法检测PBMCIL - 4和IL - 10mRNA表达。结果 :哮喘患者PBMCIL - 4mRNA的表达显著高于正常对照组 (P <0 .0 5 ) ,IL - 10mRNA的表达低于正常对照组 (P <0 .0 5 )。rIFN -γ作用于PBMC 48h后 ,哮喘患者PBMCIL - 4mRNA的表达显著降低 (P <0 .0 5 ) ,IL - 10mRNA的表达明显上升 (P <0 .0 5 ) ,而健康者PBMCIL - 4和IL - 10mRNA的表达均无显著改变 (P >0 .0 5 )。结论 :rIFN -γ可能通过下调哮喘患者PBMCIL- 4mRNA的表达、上调IL -
Objective:To study the regulation effect of rIFN-γ on expression of IL-4 and IL-10 mRNA in PBMC of asthmatic patients.Methods:PBMC were isolated by ficoll-hypaue density gradient centrifugation from 9 asthmatic patients and 5 normal persons.The cells were divided into two groups: group A(rIFN-γ,0.2 U·μl -1) and group B(control group),and then cultured in RPMI-1640 at 37℃ under 5% CO 2 for 48 h.The expression of IL-4 and IL-10 mRNA in PBMC were detected by RT-PCR.Results:The expression of IL-4 mRNA in PBMC of asthmatic patients was markedly higher than that of the control group(P<0.05)and IL-10 mRNA was significantly decreased(P<0.05). After PBMC was cultured with rIFN-γ for 48 h,the expression of IL-4 mRNA in PBMC of asthmatic patients was obviously decreased(P<0.05),and IL-10 mRNA was significantly elevated(P<0.05).There was no statistical difference observed before and after the culture with rIFN-γ in the control group(P>0.05).Conclusion:rIFN-γ can increase its anti-inflammatory function by down-regulating the expression of IL-4 mRNA and up-regulating the expression of IL-10 mRNA in PBMC of asthmatic patients.
