白细胞介素4启动子区C+33T多态性与血浆IgE水平关系 被引量:1

摘要 目的 探讨白细胞介素(IL)-4基因启动子区多态性与湖北汉族儿童哮喘的易感性及血浆IgE水平的关系。方法 用聚合酶链反应和限制片段长度多态性(PCR—RFLP)的方法对湖北汉族115例哮喘患儿及75例非哮喘对照儿童进行了+33位点多态性分析,用化学发光法测定血浆总IgE水平。结果 哮喘儿童血浆平均总IgE为351.21 IU/ml,非哮喘对照儿童血浆平均总IgE为15.48 IU/ml,有极显著性差异(P<0.0001)。哮喘患儿与非哮喘儿童IL-4基因启动子C+33T位点的等位基因频率和基因型频率CC、CT、TT之间有极显著性差异(P<0.0001),且哮喘患儿CT和TT之间均与血浆IgE升高相关(P<0.0001)。结论 IL-4启动子+33位点多态性与湖北汉族儿童哮喘易感性和血浆总IgE水平相关。 Objective To investigate the correlation between the polymorphism of IL-4 promoter region and the susceptibility of Chinese children with asthma in Hubei province and the totaJ serum IgE levels. Methods A C + 33T polymorphism in the IL-4 promoter region was determined by PCR-RFLP in Han nationality of Hubei population. 115 asthmatic children served as experimental group and 75 age-matched children without asthma were used ;as control. The total serum IgE was detected by ACS180. Results The mean total serum IgE level in experimental group and control group was 351.21 IU/ml and 15.48 IU/ml, respectively (P< 0.0001). The allele frequency of + 33 (C/T) site of IL-4 gene promoter region and genotype frequency of CC, CT, TT was significantly different between experimental group and control group (P < 0.0001). The analysis of ANOVA model suggested a genetic association of C+33T polymorphism in IL-4 promoter region with elevated total serum IgE levels (P<0.0001).Conclusion The C + 33T polymorphism in the IL-4 promoter region is associated with susceptibility of allergic asthma and elevated total serum IgE levels in Chinese children of Han nationality in Hubei province.
