摘要
应用SMARTTM技术构建了斜带石斑鱼Epinephelus coioides卵巢cDNA文库.所构建的cDNA文库滴度为4.5×106pfu/mL,重组率为99%,扩增后文库的滴度为1.2×1010 pfu/mL.把phagemid转化为质粒后,随机挑选60个阳性克隆进行酶切鉴定和测序.酶切结果表明:插入片段长度均在500~3 000bp之间.测序结果经过生物信息学分析,发现其中一个克隆为脂肪酸结合蛋白(FABP)基因全长,并与人肝型(L)脂肪酸结合蛋白同源性最高,为70%.
A cDNA library from the ovary of Epinephelus coioides was constructed using the SMART?technique. The liter of the primary library was 4.5
× 106 pfu/mL; the recombination rate was 99% ; and the titer of the amplified library was 1.2 ×1010 pfu/mL. After conversion of phagemid to plasmid, the positive clones were selected randomly and sequenced, and results showed that the sizes of the inserts were from 500 bp to 3 kb. After bioinformatic analysis, a clone of fatty acid binding protein gene was obtained, and it shared highest homology (70%) with human liver-type (L) fatty acid binding protein gene (L-FABP) .
